摘 要:目的:观察碱性成纤维细胞生长因子(bFGF)对血管平滑肌细胞(VSMC)connexin43蛋白(Cx43)表达的影响。方法:将组织贴块法培养的兔胸主动脉VSMC随机分为空白组和实验组(bFGF浓度为3μg/L),应用免疫荧光组织化学技术检测connexin43表达。用Leica激光共聚焦显微镜观察并拍照,图片用Silicon Graphics Octane进行处理。结果:免疫荧光显示,正常VSMC中Cx43蛋白主要分布于细胞中间部和周边部。bFGF作用24 h后,VSMCs数目多于对照组,Cx43蛋白荧光强度明显高于对照组,且在细胞连接处增多。结论:bFGF不仅刺激成纤维细胞的分裂增殖,对血管平滑肌细胞Cx43蛋白表达有明显促进作用,揭示间隙连接表达的增多,可能对损伤后血管重塑起一定的作用。Objective: To investigate the effects ofbFGF on expression of connexin43 in vascular smooth muscle cells (VSMC). Methods: VSMC cultures from rabbit thoracic aorta were randomly divided into experimental and control groups. The expression of connexin43 protein was detected by immunofluorescence with specific antibody against connexin43. The result was viewed and recorded with Leica laser confocal microscope. The data were processed with silicon graphics octane system Results: In control VSMC,Cx43 protein mainly located in the middle and peripheral site. The fluorescence intensity of Cx43 protein was significant higher in VSMC treated with bFGF for 24 h than that in controls. This was more obvious in the site of cell junction. In addition, treatment with bFGF also increased the number of VSMC. Conclusions: bFGF not only stimulates the division and proliferation of fibroblast cell,but also acts as modulators to promote VSMS proliferation and vascular connexin43 expression. Cx43 protein expression in smooth muscle cells,which indicates the increase of gap junction, may play certain function in vascular remodeling after injury
