摘  要:目的观察骨形成蛋白4(BMP-4)及其信号转导蛋白(Smad)5、6在去卵巢大鼠骨组织中的表达。方法采用切除大鼠双侧卵巢法复制骨质疏松模型,术后94d,用双能X线骨密度仪检测骨密度;用Western印迹法检测骨组织中BMP-4和Smad5、6蛋白表达水平。结果与正常组比较,模型空白组大鼠股骨骨密度显著下降(P<0.01);BMP-4和Smad5蛋白表达水平显著降低(P<0.01),Smad6蛋白表达水平明显升高(P<0.01)。结论骨组织中BMP-4、Smad5蛋白表达水平的下降和Smad6蛋白表达水平的升高,可能是骨质疏松发生的重要机制之一。Objective To study the expression and significance of the bone morphogenetic protein-4 (BMP-4) and signal transduction protein Smad5, 6 in the bone tissue of ovariectomized rat. Methods The osteoporosis (OP) model rat was established by removal bilateral ovaries. Bone mineral density (BMD) was measured by Dual-Energy X Ray, BMP-4, Smad5,6 protein were measured by Western blotting on 94 days after surgery. Results Compared with the normal rats, the BMD of femoral bone in vitro in model group decreased obviously ( P 〈 0. 01 ), the expressions of BMP-4 and Smad5 protein decreased obviously( P 〈 0.01 ) and the expression of Smad6 protein increased obviously ( P 〈 0. 01 ) in model group. Conclusions Conclusions The decrease of BMP-4 and Smad5 and the increase of Smad6 in bone tissue may be one of the mechanisms of postmenopausal OP.
