摘 要:目的探讨Genistein后处理对大鼠心肌缺血/再灌注损伤产生保护效应的可能机制。方法实验分7组:假手术组、缺血/再灌注组、Genistein组,ICI182780组,正钒酸钠组,Genistein+ICI182780组,Genistein+正钒酸钠组;利用Langendorff离体心脏灌流技术,建立离体大鼠心脏局部缺血/再灌注Genistein后处理模型,应用工具药物雌激素受体拮抗剂(ICI182780)和酪氨酸磷酸酶抑制剂(正钒酸钠)给予干预,观察干预前后,离体心脏局部缺血/再灌注Genistein后处理模型大鼠心肌梗死面积、心肌酶和心功能等指标变化。结果与对照组相比,缺血再灌注组增高了心肌梗死区面积,降低了心脏灌流漏出液样品中超氧化物歧化酶(SOD)活力、左室收缩压(LVSP)-左室舒张压(LVEDP)恢复率等(P<0.05)。经不同药物干预后,ICI182780组、正钒酸组、Genistein+ICI182780组的心肌梗死区面积占危险区面积的百分比高于Genistein组(P<0.05),T-SOD活力低于Genistein组(P<0.05)。结论Genistein后处理对大鼠心肌缺血/再灌注损伤所发挥的保护作用,与Genistein本身具有的雌激素特性有关;而其本身具有的酪氨酸蛋白激酶抑制作用,对心肌保护是不利的。Objective To investigate the meehanisms of cardioprotection against iscihemia-reperfusion injury in vitro with Genistein pharmacological post-treatment in rats. Methods Seven groups of rats were allocated, including the sham operated group, the ischemia-reperfusion group and five groups receiving post-treatment with Genistein, ICI 182780 vamadate, Genistein+ICI 182780, Genistein+vamadate. In vitro model of ischemia-reperfusion was established using Langendorff isolated heart perfusion techniques. Estrogen receptor antagonist (ICI 182780) and tyrosine phosphatase inhibitor (vanadate) were used as tools drug in the intervention. Differences in myocardial infarction, myocardial enzyme, heart function indexes were compared among the groups. Results Protective effect against ischemiareperfusion injury with Genistein pharulacological post-treatment was identified. This action was significantly reduced by estrogen receptor antagonist ICI 182780, and enhanced by tyrosine phosphatase inhibitor vanadate. Conclusion Protective effect against ischemia-reperfusion injury with Genistein pharmacological post-treatment may be related to its estrogen-like action. On the other hand, properties of Genistein as a tyrosine kinase inhibitor appear to compromise its myocardial protection.
