PCR-RFLP用于5种斑潜蝇的诊断和鉴别

摘　　要：在5种斑潜蝇COⅡ基因序列变异分析的基础上,建立了PCR-RFLP分子标记对该5种斑潜蝇种类的检测鉴定方法,寻找出能够检测出检疫性害虫三叶斑潜蝇及美洲斑潜蝇、南美斑潜蝇的特征酶:XbaⅠ对三叶斑潜蝇和番茄斑潜蝇均有一个酶切位点,但位于不同的酶切位点;EcoRⅠ只对美洲斑潜蝇有一个酶切位点;PVUⅡ只对南美斑潜蝇有一个酶切位点。The molecular technology of PCR - RFLP of CO Ⅱ gene was first developed to identify three species of Liriomyza in this study. Basing on PCR - RFLP, three special restriction enzymes is used for differentiating all five species of Liriomyza ： EcoR Ⅰ only digested the amplified CO Ⅱ product of L. sative ; PVU Ⅱ only for L. huidobrensis ; Xba I can digest the amplified CO Ⅱ product of L. trifolii and L. bryoniae but it cut the two species into different fragments. This paper showed that PCR - RFLP analysis of mtDNA CO Ⅱ gene was a valuable method for identification of the genus of Liriomyza.
