摘 要:目的建立气相色谱法用于测定β-榄香烯脂质体的药物含量及包封率。方法以SE-54毛细管柱(30m×0.25mm,0.33μm)为分离柱,正辛醇为内标物,无水乙醇为提取剂和定容剂,测定脂质体中药物含量。采用超滤法分离β-榄香烯脂质体中游离药物。结果该方法β-榄香烯质量浓度线性范围为0.01964~1.5712mg/mL(r=0.9992),平均回收率为97.30%,日内RSD及日间RSD均小于2%(n=5);超滤法能很好地将脂质体和游离药物分离,游离药物平均回收率为96.30%,脂质体不能透过超滤器。结论该方法准确可靠、简单快速,可用于β-榄香烯脂质体含量及包封率的测定。Objective To establish a ultrafilrtation- GC method for the determination of content and entrapment efficiency of β-elemene liposomes. Methods The separation was performed with SE-54 capillary column (30m×0.25mm,0.33μm), n-octanol was used as internal standard, anhydrous ethanol was used as extractant reagent, and the content of β-elemene liposomes was determined. Ultrafiltration was applied to separate free β-elemene from the liposomes. Results There was a good linear relation over the range of 0.01964 - 1.5712 mg/mL (r=0.9992), with an average recovery rate of 97.30%. Both the intra-day RSD and inter-day RSD were less than 2% (n = 5). Free β-elemene was well separated from liposomes with uhrafiher, the average recovery rate of free β-elemene was 96.30%, the liposomes could not permeate the uhrafiher. Conclusion This method is simple, accurate, sensitive and applicable for the determination of content and entrapment efficiency of β-elemene liposomes.
