摘 要:运用PCR-SSCP和DNA测序的方法检测高邮鸭Ghrelin基因多态性,分析其对产蛋性能的遗传效应。结果表明:①Ghrelin基因内含子1的157 bp处发生了9 bp缺失,发现了AA、BB、AB 3种基因型,A、B等位基因频率分别为0.24和0.76。B等位基因对产蛋数、开产体重表现为正效应,BB、AB基因型个体的产蛋数显著高于AA基因型个体(P<0.05)。②内含子2的431 bp处发生了T→C的突变,发现了CC、DD、CD 3种基因型,C、D等位基因频率分别为0.41和0.59。C等位基因对开产体重具有正效应,CD基因型个体开产体重显著高于DD基因型(P<0.05)。③外显子3的909 bp处发生了A→G的突变,同时产生了苏氨酸→丙氨酸的变化,发现了EE、FF、EF 3种基因型,E、F等位基因频率分别为0.33和0.67。F等位基因对开产体重、蛋重、最大连产天数具有正效应,EF基因型个体的最长连产天数显著高于EE、FF基因型(P<0.05)。The genetic polymorphism of Ghrelin gene was detected by PCR-SSCP and DNA se- quencing in Gaoyou duck. The genetic effects between Ghrelin gene and egg performance were analyzed. The results showed that. ①There were 9 bp deletions at 157 bp of intron 1, and the frequency of A/B allele of intron 1 was 0.24 and 0.76, respectively. The B allele gene had positive effect on egg production and body weight at first laying. BB and AB genotypes had higher egg production compared to AA genotype (P〈0.05). ②There was T→C at 431 bp site, and the frequency of C/D allele of intron 2 was 0. 41 and 0.59, respectively. The C allele gene had positive effect on body weight at first laying egg. CD genotype had higher egg production compared to DD genotype (P〈0.05). ③There was A→G at 909 bp site which caused Thr→Ala change, and E/F allele of exon 3 was 0.33 and 0. 67, respectively. The F allele gene had a positive effect on egg production and body weight at first laying, egg weight and highest clutch days. EF genotype had higher clutch days than EE and FF genotypes (P〈0.05).
