摘  要:选取采胶强度分别为严重过度、适中、严重不足的橡胶树胶乳,高速离心分离得到C-乳清,提取C-乳清中的蛋白质进行双向凝胶电泳,并通过PDQuest软件分析比较。结果表明:采胶强度严重不足的与其它采胶强度的相比,橡胶树胶乳C-乳清的差异表达蛋白质点有17个,其中,14个点相对上调,3个点相对下调;采胶强度适中的与其它采胶强度的相比,橡胶树胶乳C-乳清的差异表达蛋白点质有12个,其中,5个点相对上调,7个点相对下调;采胶严重过度的与其它采胶强度的相比,橡胶树胶乳C-乳清的差异表达蛋白质点有14个,其中,8个点相对上调,6个点相对下调;随着采胶强度的增加而明显上调的蛋白质点有1个;发现采胶强度高的橡胶树较采胶强度低的橡胶树胶乳C-乳清蛋白质下调的数量更多,上调的数量更少,并且采胶强度相差越大,下调与上调的比例也越大。通过研究不同采胶强度下的橡胶树胶乳C-乳清蛋白质差异,找出采胶强度与胶乳C-乳清蛋白质的关系,并利用这种关系进行乙烯刺激采胶强度的判断,进而指导采胶生产和研究。Latex was collected from rubber trees which were over-exploited, moderate-exploited or underexploited. The C-serum of the latex was isolated by ultra-centrifuge, and subsequently the proteins were extracted and applied to two-dimensional gel electrophoresis. The resultant data were analyzed by using the PDQuest Software. Seventeen differentially expressed protein spots in the 2-D gel of latex protein were found from the under-exploited rubber trees, of which 14 were up-regulated and 3 down-regulated; 12 differentially expressed protein spots from the moderate-exploited trees: 5 up-regulated, and 7 downregulated; and 14 differentially expressed protein spots from over-exploited trees: 8 up-regulated and 6 down-regulated. There was only one protein spot significantly up-regulated with the increase of tapping intensity. The latex from the rubber trees exploited in a higher intensity had a higher number of down- regulated C-serum protein and a lower number of C-serum protein than that from the rubber trees exploited in a lower intensity. The higher difference in exploitation intensity the higher ratio of upregulated to down-regulated C-serum protein in the latex. The differences in C-serum protein from latex collected from rubber trees under various exploitation intensities were analyzed to find the relation between the differential expressed proteins and the exploiting intensity, and this relation could be used to assess a stimulated exploitation intensity for giving an advice on tapping and exploitation research.
