Determination of baicalin and ephedrine hydrochloride in coryza dripping solution by RP-HPLC with dual wavelengths and gradient elution

摘　　要：【目的】建立RP-HPLC测定鼻炎滴剂中的黄芩苷和盐酸麻黄碱。【方法】采用双波长检测和梯度洗脱方法；色谱柱：Diamonsil^TM C18（250mm×4.6mm，5μm）；流动相：A为乙腈，B为1.0％磷酸溶液（含0.4％三乙胺），流速为1.0ml／min；检测波长：在0-7min为210nm，7-30min为280nm；柱温30℃。【结果】黄芩苷在0.64-3.02μg范围内线性关系良好，回归方程为：A=2560.2C-141.97，r=0.9997（n=6），平均回收率101.6％，RSD为0.86％。盐酸麻黄碱在0.20-0.97μg范围内线性关系良好，回归方程为：A=1188C＋277.59，r=0.9996（n=6），平均回收率100.9％，RSD为0.45％。【结论】本法灵敏、简便、准确，可用于鼻炎滴剂的质量控制。[Objective] To establish an RP-HPLC method for the assay of baicalin and ephedrine hydrochloride in coryza dripping solution. [Methods] Using dual wavelengths detection and gradient elution, the chromatographic conditions included Column Diamonsil^TM C18 （250mm×4.6mm,5μm）, mixtures of 1.0% phosphoric acid （content 0.4% triethylarnine ）-acetonitrile as mobile phase, detection wavelength were 210 nm （0-7 min） and 280 nm （7-30 min）, the flow rate was 1.0 ml/min, and the column temperature was 30℃. [Results] The linearity of baicalin was good in the range of 0.64-3.02μg, the regress equation was A=2560.2C-141.97, r = 0.9997（n=6）, and average recovery rate was 101.6% （RSD = 0.86%） . The linearity of ephedrine hydrochloride was good in the range of 0.20-0.97μg, the regress equation was A = 1188C ＋ 277.59, r = 0.9996 （n = 6）, and average recovery rate was 100.9% （RSD 0.45%） . [Conclusions] The method is sensitive, simple, accurate, and can be used for the quality control of coryza dripping solution.
