摘  要:目的:建立准确的自动生化分析测定血清胆红素的方法.方法:采用双试剂测定,以尿素、咖啡因及Triton-X100组成的加速剂作为第一试剂,重氮试剂为第二试剂测定总胆红素。以对氨基苯磺酸溶液为第一试剂,亚硝酸钠浓度为1.45mmol/L的重氮试剂为第二试剂测定直接胆红素。结果:总胆红素回收率为99.85%,批内CV为0.82%,批间CV为1.54%,与咖啡因法比较r=0.9999。t=1.2323,P>0.2,直接胆红素与改良Doumas法比较,r=0.9986,t=1.2508,P>0.2,批内CV为1.20%,批间CV为1.91%,脂肪乳糜等对本法无干扰,0.5g/LHb有4%的负干扰。试剂至少稳定三个月.结论:方法结果准确,试剂稳定,可作为常规方法使用.objertive To establish accurate automated analysis procedure for determining the serum bilirubinMethods Urea .caffeine and Trion - X 100 were used as the first reagent and diazo p - amino - benxene -sulphonic acid anhy as the second reagent for determining the total bilirubin. To measure the direct bilirubin, the p -aniobenxene -sulphonic acid anhy was used as the first reagent and 1. 45 mmol/L sodium nitrite diazo - p-aminobenxene-sulphonic acid anhy as the second reagent. Results The total bilirubin concentrations measured by this procedure. (Y) in 90 sera correlated well with those results obtained with the modify Jendrassik-Grof(X), Y=1. 144X- 3.83,r= 0. 999 9. Within-run was 0. 82% and between -run was 1. 54% respectively. The calibration curves was linear in range of 0 to 513 umol/L. The recovery rate was between 99. 7 and 101. 0%. The direct bilirubin results was correlated well with the modify Doumas' (x ) Y= 0. 982X+0. 25,r=0.998 6 (n= 90). Within -run was 1. 20% and between-run was 1. 91 % respectively. The common interference were not effected, but bemoglobn pede a reduce effect. The reagents were stable for three months. Conclusions the results determinate by this procedure were accurate and the reagents were stable. It may be used for routine method.
