二氮嗪对异丙肾上腺素诱导的大鼠心肌细胞凋亡相关基因表达的影响

摘  要:目的研究线粒体ATP敏感性钾通道开放剂二氮嗪对异丙肾上腺素诱导的大鼠心肌细胞凋亡相关基因表达的影响。方法采用完全随机化设计,将32只雄性SD大鼠随随机分为4组:A.正常对照组;B.异丙肾上腺素组;C.二氮嗪组;D.格列本脲组。采用半定量RT-PCR法检测各组大鼠心肌Bcl-2、Caspase-3相对表达量;采用TUNEL法检测各组大鼠心肌凋亡,计算凋亡指数(apopticindex,AI)。结果各组大鼠心肌Bcl-2、Caspase-3相对表达量之间存在差异(P<0.01)。C组Bcl-2、Caspase-3相对表达量与A,B,D组均有差异(P<0.05);B组、D组与A组之间有差异(P<0.05);B组、D组之间无明显差异(P=0.134,0.297)。A组大鼠心肌TUNEL染色未见阳性细胞;B、B、D各组AI上存在差异(P=0.001);C组和B、D组织间存在差异(P<0.01);B组、D组之间无明显差异(P=0.156)。结论二氮嗪可以减少异丙肾上腺素诱导的大鼠心肌细胞凋亡;减少凋亡的原因可能与其能上调Bcl-2的表达,下调Caspase-3表达有关。Objective To research the influence of diazoxide to apoptotic gene expression of rat myocardial cell induced by isoprenaline. Methods 32 male rats (Sprague-Dawley) were allocated to four groups randomly, A. Normal group(control group); B. Isoprenaline group (to induce cell apoptosis); C. Diazoxide group (mitoKATP channel opener); D. Glibenclamide group (mitoKATP channel inhibitor).The relative quantity of Bcl-2 and caspase-3 expression of each group were measured by RT-PCR(reverse transcription polymerase chain reaction) technique. Myocardium cell apoptosis of each group was tested by TUNEL(terminal-deoxynucleotidyl transferase mediated nick end labeling) technique and the apoptotic index (AI) was calculated. Results The relative quantity of Bcl 2 expression of C group is higher than Iso group and D group (P〈0.05).But the relative quantity of Caspase-3 expression of C group is lower than B group and D group (P〈0.05). There is no significant difference on the relative quantity of Bcl-2 and Caspase-3 expression between B group and D group. The P value is 0.134 and 0.297 respectively. The AI of C group is lower the B group and D group (P〈0.01). The AI between B group and D group has no significant difference (P=0.156). Conclusion The diazoxide can protect myocardium cell from apoptosis induced by isoprenaline. The mechanism may be that diazoxide can up-regulation the Bcl-2 expression and down-regulation the caspase 3 expression.
