分泌抗IBDV单抗的杂交瘤细胞在新型5L细胞反应器中连续培养的研究

摘　　要：能稳定分泌抗ＩＢＤＶ单抗的杂交瘤细胞，直接从方瓶中接种进入已改装的新型５Ｌ细胞反应器，接种浓度不少于３×１０＾８个细胞／Ｌ，细胞在ｐＨ７，１０，ＤＯ５５，５０－７０ｒ／ｍｉｎ，３７℃的培养条件下生长良好；在保持反应器内的葡萄糖浓度不低于１．０ｇ／Ｌ的情况下，平均每天可生产单抗３Ｌ左右，产物的平均ＯＤ值为０．３６８，持续周期达３０ｄ左右。Hybridoma cells which could stably secrete monoclonal antibody to IBDV were inoculated from T-flasks in the new type 5L Celligen with cell density more than 3×108/L,The hybridoma cells grew well under the culture condition of pH7.10, DO55,50-70rpm, 37℃ ;about 3L McAb could be produced every day when the concentration of glucose in Celligen was sustained at 1.0g/L.The of the harvest was 0.368, the cultivation cycle was about 30 days.
