摘  要:【目的】通过观察穴位埋线对更年期大鼠下丘脑去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)等单胺类神经递质的影响,探讨其对更年期综合征防治作用的机理。【方法】选用60只SD雌性大鼠,随机分为治疗组、模型组和假手术组各20只;除假手术组外,其他组均采用手术摘除大鼠双侧卵巢复制去势更年期大鼠模型,治疗组选取肾俞、三阴交、子宫穴进行穴位埋线,1次/周,连续4周;采用放射免疫法检测下丘脑组织NE、DA、5-HT、5-HIAA含量。【结果】模型组较假手术组的NE、DA水平显著降低(P<0.05或P<0.01),5-HT、5-HIAA水平显著上升(P<0.01或P<0.001),5-HT/NE比值显著升高(P<0.001);穴位埋线组与模型组比较,可显著升高衰退的NE水平(P<0.05),降低5-HT、5-HIAA水平(P<0.05或P<0.01),且埋线组的5-HT/NE比值亦较模型组显著降低(P<0.01)。【结论】穴位埋线治疗更年期综合征的作用可能与其能调整去势雌性大鼠下丘脑单胺类神经递质,纠正下丘脑紊乱的植物神经功能有关。Objective To investigate the effects of acupoint catgut-embedding therapy(ACT) on hypothalamic of hypothalamic norepinephrine(NE),dopamine(DA),5-hydroxytryptamine(5-HT),5-hydroxyindole acetic acid(5-HIAA) in menopause rats,and to explore the preventive and therapeutic mechanism of ACT for climacteric syndrome.Methods Sixty female SD rats were equally randomized into treatment group,model group and sham-operation group.Except the rats in the sham-operation group,the rats in other groups received removal of bilateral ovaries to establish ovariectomized menopause rat models.The treatment group received catgut-embedding on acupoints of Shenshu,Sanyinjiao and Zigong,once per week,and the treatment lasted 4 weeks.After treatment,the hypothalamic NE,DA,5-HT,5-HIAA levels were measured by radioimmunoassay.Results NE and DA levels were significantly decreased(P〈0.05 or P〈0.01),5-HT and 5-HIAA levels were increased(P〈0.01 or P〈0.001) and the ratio of 5-HT/NE was significantly increased(P〈0.001)in the model group as compared with those in the sham-operation group.Compared with the model group,NE was increased(P〈0.05),5-HT and 5-HIAA levels and the ratio of 5-HT/NE were decreased in the treatment group(P〈0.05 or P〈0.01).Conclusion The therapeutic mechanism of acupoint catgut-embedding therapy for climacteric syndrome is probably related with the regulation of hypothalamic monoamine neurotransmitters and with the correction of hypothalamus autonomic function disorder.
