摘 要:用薄膜分散法制取荧光红GG脂质体,紫外分光光度计520 nm处测其吸光度,计算总浓度.取荧光红GG脂质体,3000 r/min,离心10 min,分离含药脂质体和游离的药物,稀释后测定其上清和沉淀吸光度,计算其浓度.由包封率=(总浓度-沉淀浓度)/总浓度×100%计算包封率.结果发现溶液的稳定性和精密度都良好,测得脂质体中药物的包封率分别为11.89%,11.46%,12.89%,结果较为接近,说明低速离心法是一种有效的水不溶性药物脂质体包封率的测定方法.The fluorescence red GG liposomes was prepared by thin film dispersed method. The free drugs and drugs entrapped in liposomes was separated by centrifugating 10 min at the speed of 3 000 r/min. The drug concentration was determined by UV spectrophotometry at 520 nm and the entrapment efficiency was calculated. The good linearity, recoveries, stability and precision showed that the Ultraviolet Spectrophotometry was credible. The entrapment efficiency of fluorescence red GG in liposomes was 11. 89%, 11. 46% and 12. 89%, respectively. All the results suggest that the low-speed centrifugation is suitable for determining the entrapment efficiency of water-insoluble drugs in liposomes.
