摘 要:目的阐述引起安徽省2008年手足口病(hand,foot and mouth disease,HFMD)流行的肠道病毒71型(enterovirus71,EV71)基因特征。方法采集HFMD暴发初期患者标本,进行病毒分离、培养,通过逆转录-聚合酶链反应(RT-PCR)鉴定上述培养物。扩增病毒株及核酸样本的VP1编码区全基因,并进行序列测定和分析。结果暴发初期分离出3份病毒株,经中和试验及RT-PCR证实为EV71。根据VP1全基因序列与EV71基因型、亚型参考株构建亲缘性关系树,17份测序结果与C4亚型聚为一簇,与A、B、C1、C2、C3、C4基因型别(亚型)的核苷酸的同源性分别为:82.0%-83.0%、84.3%-85.4%、89.7%-91.1%、89.1%-90.7%、88.3%-89.8%、93.0%-94.6%;氨基酸的同源性分别为:93.9%-95.2%、96.6%-97.6%、97.9%-98.6%、98.6%-99.3%、98.3%-98.9%、98.6%-99.6%;簇内比较显示,1株2006年的EV71毒株与另16份2008年样本在C4亚型内又分属两个亚簇,核苷酸、氨基酸的同源性分别为:96.2%-96.9%、98.3%-99.3%。结论引起安徽省2008年HFMD的EV71为C4亚型,与2006年分离的1份毒株有差异,提示安徽省存在C4亚型的不同分支。Objective To reveal the genetic characteristic of enterovirus 71 (EV71) strains isolated during an epidemic of Hand-Foot-Mouth Disease (HFMD) in Anhui, 2008. Methods Virus were isolated from patients in early outbreak of HFMD. The EV71 strains were identified by microneutralization assay and reverse transcriptase PCR (RT-PCR). The VP1 complete gene of strains and nucleic acid samples were amplified and sequenced. Then the sequences were analyzed by software. Results Three EV71 strains were isolated in early outbreak. Data from microneutralization and RT-PCR indicated that all the strains belong to EV71. The phylogenetic tree was constructed between isolated and other EV71 reference strains based on alignment of the partial sequence of VP1 gene. All the results clustered with reference strains of CA subgenotype in the phylogenetic tree. The nucleotide homogeneity of 17 samples with the representative isolates of A, B, C1, C2, C3 and CA genotypes(subgenotype) were 82.0 % 83.0%, 84.3%-85.4%, 89.7%-91.1%, 89.1%-90.7%, 88.3%-89.8%, 93.0%-94.6% and the amino acid homogeneity were 93.9%--95.2%, 96.6%--97.6%, 97.9%--98.6%, 98.6% -99.3%, 98.3%--98.9%, 98.6% -99.6% respectively. In CA subgenotype, the strain of 2006 and other 16 samples of 2008 belong to two clades , the nucleotide and amino acid homogeneity of two clades were 96.2 % -- 96.9 % and 98.3 % - 99.3 %. Conclusions The EV71 strains caused HFMD in Anhui belonged to CA subgenotype, but strains of 2008 differs from one strain of 2006 in C4 subgenotype, suggesting that strains of CA subgenotype in Anhui may have different clades.
