降脂复肝汤对非酒精性脂肪肝大鼠肝脏的保护作用

摘　　要：目的观察降脂复肝汤对高脂饮食诱导的非酒精性脂肪肝大鼠肝脏的保护作用。方法用高脂饮食建立大鼠非酒精性脂肪肝模型。用降脂复肝汤药物治疗12周,测大鼠体重、肝湿重分析肝指数;通过肝脏组织HE染色观察肝组织病理学、肝功酶试剂盒及血脂试剂盒检测血清生化指标,以及免疫组织化学检测白细胞介素1α(IL-1α)的变化。结果降脂复肝汤组大鼠肝指数、血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(A ST)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、肝组织TG均较模型组显著降低(P<0.05或P<0.01)。治疗降脂复肝汤组IL-1α的表达较模型组明显降低(P<0.05)。结论降脂复肝汤能明显改善高脂饮食诱导的脂肪肝大鼠肝脏的病理学组织变化及肝组织脂肪变性,也能降低脂肪肝变性肝组织IL-1α的表达,这可能是其干预高脂饮食诱导的非酒精性脂肪肝大鼠的作用机制之一。Objective To observe the protective effect of Jiangzhi Fugan Decoction （Decoction for lipid-lowering and recovery of hepatic function） on the hepatic function in rats with non-alcohol fatty liver induced by high fat feeding . Methods Rat models with non-alcohol fatty liver were established by feeding high fat food. They were treated with Jiangzhi Fugan Decoction for 12 weeks. Then, the body weight and liver wet weight were measured to analyze the liver index, the pathological examination of liver tissues was performed under a microscope by HE staining, the biochemical indices of liver function and blood lipid were measured, and the level of IL-1α in hepatic tissue was detected by immunohistochemical method. Results The liver index, serum levels of alanine transaminase （ALT）, aspartie transaminase （AST）, triglyceride （TG）, low density lipoprotein cholesterol （LDL-C）, and hepatic tissue TG decreased in Jiangzhi Fugan Decoction group （P〈 0. 05 or P〈0.01） as compared with those of the model group, and immunohistochemical examination showed decrease of IL-1α expression in the hepatic tissue as well （P〈0.05）. Conclusion Jiangzhi Fugan Decoction is effective in improving the pathological changes in hepatic tissues and steatosis, at the same time, in decreasing IL-1α expression. This is might be its mechanism in treating non-alcohol fatty liver induced by high fat feeding in rats.
