摘 要:目的建立猫豆中酪氨酸含量的测定方法。方法采用高效液相色谱法,色谱柱为Aichrombond-AQC18柱(250mm×4.6mm,5μm),流动相为8.5mmol/LNaAc溶液(磷酸调节pH=4.0)-甲醇溶液(99:1,V/V),检测波长为230nm,流速为1.0mL/min。结果酪氨酸进样质量浓度在7.5~17.5μg/mL范围内与峰面积积分值线性关系良好,r=0.9995(n=5),平均回收率为100.39%,RSD=0.86%(n=6)。结论该方法测定简便、快速、灵敏、重现性好,可用于测定猫豆中酪氨酸的含量。Objective To establish a HPLC method for the determination of tyrosine in Stizolobium cochinchinensis (Lour.) Tang et Wang. Methods To determine the tyrosine content by HPLC. The Aichrombond-AQ C18 column(250 mm ×4. 6 mm, 5 μm)was used. The mobile phase was 8.5 mmol/L sodium acetate solution(phosphate regulation pH=4.0)-methanol solution(99 : 1, V/V).The detection wavelength was 230 nm. The flow rate was 1.0 mL/min. Results The linear range of calibration curve was 7.5 - 17.5μg/mL for tyrosine (r =0. 999 g, n = 5). The average recovery rate was 100. 39% and RSD was 0. 86% ( n = 6). Conclusion The method is fast, simple, sensitive and stable. It has been proved to be a good practice for the determination of tyrosine from Stizolobium cochinchinensis (Lour.) Tang et Wang.
