滋阴泻火中药对青春期大鼠子宫与卵巢ER、IGF-Ⅰ、AR基因及蛋白表达的影响

摘　　要：目的:从基因及蛋白水平研究滋阴泻火中药对青春期大鼠子宫、卵巢雌激素受体(ER)、胰岛素生长因子-Ⅰ(IGF-Ⅰ)、芳香化酶(AR)表达的影响,从雌激素和胰岛素生长因子调控及雌激素合成酶途径探讨滋阴泻火中药防治性早熟的机制。方法:将动物分为对照组和中药组,中药组喂饲滋阴泻火中药每次3ml,每日1次,对照组仅给予等量的0.9%NaCl溶液;末次给药24h后剥离子宫称重,并采用实时荧光定量聚合酶链式反应(real-time fluorescence quantitative PCR,FQ PCR)法观察子宫、卵巢ER、IGF-Ⅰ、AR mR-NA的表达,采用免疫组化法观察子宫、卵巢ER、IGF-Ⅰ、AR蛋白的表达。结果:给予滋阴泻火中药干预后,青春期大鼠子宫重量、子宫/体质量比下降,子宫、卵巢ER、IGF-Ⅰ、AR的mRNA和蛋白的表达量均明显下调。结论:滋阴泻火中药能够显著下调靶器官雌激素受体、胰岛素样生长因子及雌激素合成酶(芳香化酶),这可能是其治疗儿童性早熟的作用机制之一。Objective： To study the effects of Chinese herbal medicine （CHM） for nourishing yin and purging fire on mRNA and protein expression of estrogen receptor（ E R）, insulin growth factor-I （IGF-I ） and aromatase （AR） in uterine and ovary of adolescent rats, for investigating the mechanism of its preventing and treating precocious puberty by means of regulating ER, IGF-I , AR mRNA and protein expression. Methods： Rats were divided into two groups ： control group and CHM group. The rats in CHM group were fed with CHM for nourishing yin and purging fire 3 ml/d, and the rats in control group was fed with normal saline of the same dosage. All rats were sacrificed 24 hours after the last administration of drugs ; the uterine wet weight, uterine/body weight were measured. Expression of ER, IGF- I and AR in the uterine and ovary were determined by real-time fluorescence quantitative PCR （FQ PCR）, expression of their protein was determined by immunohistochemistry. Results： With the interference of CHM, the uterine wet weight, uterine/body weight decreased and the expressions of mRNA and protein of ER, IGF- I and AR were down-regulated significantly. Conclusion： CHM for nourishing yin and purging fire can significantly down-regulate the mRNA and protein expression of uterine and ovary ER, IGF- I and AR, which may be one of the mechanisms of preventing and treating precocious puberty.
