摘 要:目的:对《中国药典》(2005版)中注射用苄星青霉素中青霉素含量测定的方法进行改进。方法:采用KromasilC18柱(7μm,250mm×60mm);流动相:0.05mol/L磷酸二氢钾缓冲液(用氢氧化钠溶液调节pH值至6.5)-乙腈(75∶25);流速:1.0mL/min;检测波长:225nm;柱温:35℃。结果:在该色谱条件下,青霉素峰能与其他杂质峰获得完全分离,青霉素质量浓度在0.125~2.000mg/mL范围内,峰面积响应值(A)与青霉素质量浓度(C)间有良好的线性关系(r=0.9999),平均回收率:99.60%。结论:该方法简单、快速、准确地测定注射用苄星青霉素中青霉素的含量。Objective: To improve the Chinese Pharmacopoeia (2005 edition) method for the determination of benzathine benzylpenicillin for injection. Methods: The column: Kromasil C18 (7 μm, 250 mm× 60 mm). The mobile phase is: acetonitrile-0.05 mol/L potassium dihydrogen phosphate buffer solution(25 : 75, adjusting pH to 6.5 with sodium hydroxide solution). The flow rate is: 1.0 mL/min. The detection wavelength is: 225 nm. The column temperature is: 35 ℃. Results: Benzylpenicillin could be completely separated from the other impurities. The linear range of benzylpenicillin was 0.125-2.000 mg/mL, r = 0.999 9, and the average recovery rate is 99.60%. Conclusion: The proposed method is convenient and accurate. The method can be used for the determination of benzathine benzylpenicillin for injection.
