摘 要:目的探讨胆管上皮细胞缺血再灌注损伤的机制及三七总皂苷(panax notoginseng saponins,PNS)在大鼠肝移植中胆管细胞I/R损伤中的作用。方法采用吻合肝动脉的大鼠原位肝移植模型。分三组观察在不同条件下胆管细胞的病理形态变化及PNS对此缺血再灌注损伤的影响。结果缺血再灌注的胆管细胞损伤更重,氧自由基生成更多,NF-κB表达更多,PNS预处理保护组胆管细胞损伤明显减轻。结论缺血再灌注损伤能加重胆管细胞的损伤,PNS预处理可减轻该损伤,其机制可能为抑制氧自由基生成,减少NF-κB的表达。Objective To explore the mechanisms of ischemia-reperfusion(I/R)injury of the bile duct and the effect of panax notoginseng saponins(PNS).Methods The liver transplantation model of rats was established.Pathological changes of the bile duct cells and the effective protection of PNS against I/R injury were observed and compared among three groups.Results The I/R bile duct cells has more serious injury,more oxygen free radica and more expression of nuclear factor kappa B(NF-κB).The injury is significant released in PNS pretreatment group. Conclusion I/R can worsen the injury of bile duct cells. PNS pretreatment can effectively protect against liver I/R injury. The mechanisms may be the depression of oxygen free radical production and the decrease of NF-κB expression.
