真菌菌株sf1760农用活性物质的分离、纯化及LC/MS分析

摘  要:采用液体发酵,冷冻干燥,萃取,HPLC半制备,生物测定,HPLC纯化,活性物质的LC/MS/MS分析研究真菌菌株sf1760发酵活性物质的主要成分。结果从真菌菌株sf1760中提取得到两个组分具有抑制作物病原真菌的作用,经LC/MS/MS分析组分1的紫外吸收峰为264nm,其母离子ESI+316.6,ESI-314.8;其子离子ESI+263.6,ESI-261.7,与Gliotoxin的紫外和质谱信息相同。组分2的紫外吸收峰为210nm,其质谱ESI+444.8,子离子为ESI+228.6,ESI+199.6;ESI-442.9,子离子为ESI-240.6,与Fumiquinazo-line C的紫外和质谱信息相同。The isolation procedure was guided by bioassay using phytopathogenic fungi to investigate agro-active metabolites of fungus strain sf1760. Various means of chromatography including liquid fermentation, lyophillization, extraction, semi-prep HPLC ,bioassay and preparative HPLC purification were conducted. Two compounds were isolated from the metabolites of the sf1760, and they were identified as gliotoxin,fumiquinazoline C which almost completely inhabited phytopathy,UV adsorption wavelength of compound 1 was 264 nm, ESI+316.6,ESI-314.8, the daughters were ESI+263.6,ESI-261.7,and the UV adsorption wavelength of compound 2 was 210 nm, ESI+444.8,ESI-442.9, the daughters were ESI+228.6, ESI+199.6, ESI-240.6 respectively. They were similar to gliotoxin and fumiquinazoline C according to related documents.
