磷脂对大鼠离体肠管吸收葛根素的影响

摘　　要：目的探讨磷脂（Ｐｈｏｓｐｈｏｌｉｐｉｄｓ，Ｐ）对大鼠离体肠管吸收葛根素（Ｐｕｅｒａｒｉｎ，Ｐｕｒ）的影响。方法在改良Ｆｉｓｈｅｒ－Ｐａｒｓｏｎｓ灌流的大鼠肠管标本中，以ＨＰＬＣ法测定灌流液中Ｐｕｒ的含量。结果Ｐｕｒ的吸收峰在２５４ｎｍ处，Ｐｕｒ的标准曲线呈正相关（ｒ＝０．９９６，Ｐ＜０．０１），标准品的回收率为１００．７±４．５３％，变异系数为３．２５％，在５０ｍｉｎ时，乳化葛根素和葛根素的扩散常数分别为３×１０－７和１×１０－７ｃｍ２·ｓ－１。结论Ｐ能增加Ｐｕｒ的吸收量。Objective The effect of Phospholipids(P) on the absorbtive content of Puerarin ( Pur) in isolated intestine of rats were investigated. Method At improved Fisher Parsons adevice,the content of Pur and emulsive Pur(EPur) were respectively measured by HPLC.Result The absorbtive peak of wave length of Puerarin was at 250 nm by HPLC.The standard curve of Pur appeared positive interrelating(r=0.996,P<0.01),the standard recovery was 100.7±44.52%, the confficient constant ( cv) was 3. 25%. The content of Pur and ePur were respectively determinated by the method of HPLC in isolated intestine of rats. The diffuse constant D are 1×10 -7 and 3×10 -7 cm 2·s -1 .Conclution The results show that phospholipid can promite absorbtion of Puerarin.
