掌叶半夏总蛋白诱导人卵巢癌SKOV3细胞凋亡的实验研究

摘　　要：目的研究掌叶半夏蛋白对人卵巢癌细胞SKOV3体外增殖凋亡活性的影响,以确定掌叶半夏蛋白能否抑制卵巢癌的生长及其可能的作用机制。方法应用CCK-8法检测掌叶半夏蛋白对SKOV3细胞体外生长的影响;利用流式细胞仪,采用Anexinn-V法检测掌叶半夏蛋白诱导SKOV3细胞凋亡的作用。结果SKOV3细胞经掌叶半夏蛋白作用后,细胞生长受到明显的抑制,Anexinn-V法检测到细胞凋亡,凋亡率随着作用浓度的增加及作用时间的延长而升高。结论掌叶半夏蛋白对卵巢癌细胞SKOV3有抑制增殖和促凋亡作用,其诱导凋亡机制有待进一步研究。OBJECTIVE To investigate the effects of the protein of Pinellia pedatisecta Schott on the proliferation of human ovarian cancer cell line SKOV3 in vitro and to determine whether the protein of Pinellia pedatisecta Schott inhibit the growth of ovarian cancer and its mechanism of action. METHODS CCK-8 was usedto detect the effect of the protein of Pinellia pedatisecta Schott on the growth of SKOV3 cells in vitro; using flow cytometry, detected the apoptosis of SKOV3 cells treated with the protein of Pinellia pedatisecta Schott by Anexinn-V. RESULTS The protein of Pinellia pedatisecta Schott had some effect of inhibiting the proliferation and inducing apoptosis of SKOV3 cells, and the mechanism would be further studied. CONCLUSION The protein of pinellia pedatisecta schott had some effect of inhibiting the proliferation and inducing apoptosis of SKOV3 ceils, and the mechanism would be further studied.
