摘  要:探讨不同周期性应变对骨髓间充质干细胞(mesenchymal cells,MSCs)细胞形态变化的影响。1%、3%、10%、15%拉伸应变(频率均为0.5 Hz)分别作用D1细胞(间充质干细胞系)12 h;10μg/ml细胞松弛素B(Cytochalasin B,CB)作用细胞1 h后,设相同的条件进行平行实验;CB作用细胞1 h后,10%应变作用细胞12 h和24 h,无CB细胞为对照,以上实验以非应变细胞做为对照。FITC-鬼笔环肽(FITC-Phalloidin)标记细胞微丝,碘化丙啶(propidium iodide,PI)标记细胞核,激光共聚焦显微镜观察细胞形态结构。1%、3%、10%、15%应变作用细胞后,细胞排列同受力方向分别呈平行、垂直、45°和平行排列;细胞微丝破坏后,细胞排列方式同非CB组相比有明显差异,细胞微丝结构发生断裂、重排。说明不同周期性应变能引起细胞形态结构改变,细胞形态的变化是细胞对应变反应的响应。To study the effects of different strain on morphologic change of mesenchymal stem cells(MSCs).The D1 cell line were cultured in vitro and then exposed to 1 %,3 %,10 %,and 15 % strained respectively for 12 h.Cells were treated with Cytochalasin B(CB) for 1 h,the same experiment was undertaken.The other experiments were to treat cells with Cytochalasin B(CB) for 1 h,and expose to 10% strained for 12 and 24 h,unstrained cells as control,frequency was 0.5 Hz in all experiments.Cellular microfilament were marked with FITC-Phalloidin,Propidium Iodide(PI) marked nucleus.Application of technology of Confocal Lasar Scanning Microscopy(CLSM) to observe cellular Morphologic change.Cellular arrangement were parallel,perpendicularity,45°and parallel in corresponding with forced direction in 1%,3%,10%,15% strain.After cellular microfilament were damaged by CB,cellular arrangement were obviously different to compared with strained.Cells were affected with 10% strain for 12 h and 24 h,cellular microfilament were rearranged and interrupted.The different strain can affect morphologic change of cells,it is different effect of cells exposed by strain.
