摘  要:目的:通过在Allen法基础上,采用自制击打器建立了不同程度急性脊髓损伤动物模型,并对其进行蛋白质组学研究。方法:在原有技术基础上,以IPG等电聚焦为第一向、垂直SDS PAGE为第二向,对脊髓蛋白质样品提取、等电聚焦上样量、等电聚焦电泳参数、SDS PAGE、SDS胶染色及保存进行了研究和实验条件优化,建立和优化了脊髓蛋白质组分析的双向电泳的技术。结果:本实验在国内首先成功建立了正常和损伤脊髓蛋白质组双向电泳图谱,该图谱能够从蛋白质水平上直接反映脊髓损伤所引发的脊髓蛋白质组变化的全貌。并识别和鉴定出部分差异表达的蛋白质,为阐明急性脊髓损伤后损伤和修复的机制奠立了初步基础,提供了实验依据。结论:应用改良Allen氏法成功建立了不同程度急性脊髓损伤动物模型。建立和优化了脊髓蛋白质组分析的双向电泳的技术,成功建立了分辨率高、重复性好的正常和损伤脊髓蛋白质组双向电泳图谱。Objertive:The animal model of SCI in rabbits was established with a modified Allen's method. Then we could study to Proteomics of spinal cord. Methods: In this study, the animal model of SCI on different degree of impact established with a new weight--drop device has a stable and repetitive characters for the research on spinal cord injury. Well--resolved and reproducible 2--DE technological, patterns of spinal cord were established. The 2--DE map of spinal cord of SCI might be obtained and particular differential protein could be characterized. Results:The results provide fundamental information for the molecularmechanism of SCI. Conclusion: In this study, the animal model of SCI on different degree of impact established with a new weight--drop device had a stable and repetitive characters for the research on spinal cord injury. Well--resolved and reproducible 2--DE technological. The 2--DE map of spinal cord of SCI should be obtained and particular differential protein could be characterized.
