肾气丸“阴中求阳”配伍对肾阳虚大鼠下丘脑多巴胺及五羟色胺含量的影响

摘　　要：目的用肾气丸各拆方组水煎液灌胃肾阳虚大鼠,测定灌胃后各组大鼠下丘脑中多巴胺(DA)、五羟色胺(5-HT)的含量,分析肾气丸"阴中求阳"治疗肾阳虚证的机理。方法以氢化可的松25 mg/(kg.d)注射大鼠,连续12 d,同时以各肾气丸拆方组水煎液灌胃,于灌胃第21天,麻醉大鼠,取出下丘脑,称重,以5%高氯酸匀浆,高速低温离心后分取上清液,用高效液相色谱法检测DA,5-HT的含量。结果与正常组比较,模型组大鼠下丘脑中DA、5-HT含量升高,经灌胃给药后,各拆方组大鼠下丘脑中DA,5-HT的含量均有不同程度的降低,以阴阳双补组含量最低,而补阳组降低不明显,其中阴阳双补1组含量低于补阳1组,阴阳双补2组含量低于补阳2组,具有统计学意义。结论降低下丘脑中多巴胺、五羟色胺的含量,可能是肾气丸"阴中求阳"配伍治疗肾阳虚证的机理之一。Objective To study the influence of compatibility of Shenqi Pill ＂Reinforcing Yang from Yin＂ on the contents of DA,5-HT in hypothalamus of deficient kidney-yang Rats.Methods The rats were injected with cortisone at the dose of 25 mg/（kg·d） for consecutive 12 days and adminstered with dismantled decoction at the same time.On the 21st day,the rats were sacrificed and the contents of DA,5-HT in hypothalamus were measured by the method of high performance liquid chromatogram.Results Compared with normal group,the contents of DA,5-HT was increased in model group.After intragastric administration,the content of DA,5-HT in each dismantled group were decreased at different level in hypothalamus.The first group of invigorating yin-yang was higher than the first group of invigorating yang,the second group of invigorating yin-yang was higher than the second group of invigorating yang,and all of these had statistic significance.Conclusion The decrease of DA,5-HT contents in hypothalamus is likely to be the mechanism by which Shenqi Pill treats the kidney-yang asthenia syndrome by the compatibility of ＂reinforcing yang from yin＂.
