摘  要:目的优选黄连生药中盐酸小檗碱的最佳提取工艺。方法以水作为提取溶剂,通过正交实验,高效液相色谱法测定盐酸小檗碱的含量为指标,考察提取溶剂体积、提取时间以及提取次数对提取效果的影响,优选出最佳的提取工艺。结果黄连生药中盐酸小檗碱的最佳提取工艺为药材加入8倍量水,回流提取2次,1.5h/次,测得盐酸小檗碱含量为6.5499%。结论该提取工艺条件科学,成本较低,提取得到的盐酸小檗碱含量较高,具有一定的实用价值。Objective To optimize the of extract process of berberine hydrochloride in Rhizoma Copotidis by L9(34) orthogonal method.Methods With water as extraction solvent,the contents of berberine hydrochloride determined by HPLC as the index,the effects of volume of extraction solvent,time of refluxing and extraction times were investigated and the L9(34) orthogonal experiment was adopted to optimize the process.Results The optimum extraction process was achieved with addition of water as extraction solvent(8 times raw herb in volume) into materials,refluxing 1.5hours and extracting for 2 times.The contents of berberine hydrochloride was 6.5499%.Conclusion The optimum extraction process for berberine hydrochloride is scientific,cost is comparatively low,the contents of berberine hydrochloride is high.This method has wider practical value in production.
