摘 要:目的从鼠肺中获得汉坦病毒分离株并进行基因鉴定与分型,为地区分离株特性研究提供基础。方法用Vero-E6细胞分离病毒,进行荧光和RT-PCR鉴定,之后对目的片段进行序列测定和分析。结果从褐家鼠鼠肺中分离到一株汉坦病毒,编号SC106。用汉城型(SEO)汉坦病毒M基因特异性引物检测得到预计大小的片段,此片段的核苷酸序列分析表明,SC106与SEO型参考毒株的同源性较高,均在94%以上;与汉滩型(HTN)参考毒株的同源性较低,均在67%以下。进化树分析表明SC106位于SEO型所在的支系。结论SC106为SEO型汉坦病毒。Objective To isolate and identify hantavims from rodent lung sample, and to determine the genotype of the virus. Methods Hantavirus was isolated by using tissue culture method, and was identified by using RT-PCR. Then, the obtained fragment was sequenced and analyzed. Results A strain of hantovirus was isolated from Rattus norvegien lung sample and was designated SC106. An expected M gene fragment of SC106 was ampli- fied with Seoul (SEO) virus speeitle primers. Nueleotide sequence comparison of the fragment showed that the homologies were more than 94% to SEO roferenee viruses, but less than 67% to HTN reference viruses. Phylogenefie analysis showed that SC106 ehstered in the group of SEO viruses. Conclusion SC106 most closely related to SEO virus.
