灯盏花素对单侧输尿管梗阻大鼠肾间质纤维化的保护作用

摘　　要：目的观察灯盏花素对单侧输尿管梗阻(UUO)大鼠肾脏间质纤维化的保护作用及机制。方法将72只Wistar大鼠随机均分为假手术组、UUO组和灯盏花素组。后两组大鼠结扎并剪断左侧输尿管制作UUO模型,假手术组仅游离左侧输尿管后缝合。灯盏花素组于手术前1d至处死当天给予灯盏花素注射液[100mg/(kg.d)]腹腔注射(2.5ml/次,2次/d)。假手术组及UUO组用同体积生理盐水腹腔注射。采用HE染色观察术后4、7、14d肾脏组织的病理改变,免疫组化及Western blotting检测转化生长因子β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)在肾脏的表达。结果假手术组的各时间点肾组织未见病理改变。UUO组术后4、7d,肾间质相对面积增加,且有炎性细胞浸润;术后14d可见肾小管明显扩张,肾间质明显纤维化。与同时间点的UUO组相比,灯盏花素组肾间质相对面积明显下降。与假手术组比较,UUO组和灯盏花素组的TGF-β1和α-SMA蛋白表达水平均有明显增加(P<0.01),但灯盏花素组的表达低于UUO组(P<0.05)。结论灯盏花素可以通过下调TGF-β及α-SMA的表达减轻UUO术后肾脏间质纤维化。Objective To explore the protective effects of breviscapine on the renal interstitial fibrosis induced by unilateral ureteral obstruction（UUO） in rats,and to explore its underlying mechanism.Methods Seventy-two rats were randomly divided into 3 groups（24 each）：sham operation group（sham group）,UUO group and breviscapine treatment group.Left ureters of the rats in both UUO and breviscapine treatment group were ligated and then divided to reproduce UUO model,while in sham group the ureter was isolated only without further manipulation.Breviscapine [100mg/（kg·d）] was intraperitoneally administratered to the rats in breviscapine treatment group（2.5ml once,twice a day） one day before operation till the day when rats were sacrificed.Rats in sham group and UUO group were given same amount of normal saline by intraperitoneal route.HE staining was performed for the observation of the pathological changes in kidney tissues at day 4,7 and 14 after operation.The expressions of TGF-β1 and α-SMA in kidney were detected by immunohistochemistry and Western blotting.Results No pathological change was observed in kidney tissues at every time point in sham group.On day 7 and day 14,the relative area of renal interstitium was increased and inflammatory cells infiltration was observed.Dilatation of kidney tubules and interstitial fibrosis were seen on day 14 in UUO group.The relative area of renal interstitium was smaller obviously in breviscapine treatment group compared with that in UUO group at the same time point.In comparison to the sham group,the expressions of TGF-β1 and α-SMA were enhanced significantly in both UUO and breviscapine treatment group（P〈0.01）.The expressions of TGF-β1 and α-SMA were lower in breviscapine treatment group than in UUO group（P〈0.05）.Conclusion The renal interstitial fibrosis induced by UUO may be alleviated by breviscapine through down-regulating the TGF-β1 and α-SMA expressions.
