摘  要:目的探讨高氧致新生大鼠慢性肺疾病(CLD)发生时Ⅱ型肺泡上皮细胞(AEC-Ⅱ)损伤程度,研究高氧对AEC-Ⅱ数量及功能的影响。方法建立高氧诱导新生鼠CLD模型,80只Wistar新生鼠被随机分为实验组及对照组,透射电镜观察肺组织的超微结构,免疫组化方法、RT-PCR半定量方法检测肺组织表面活性蛋白C(SP-C)蛋白及mRNA表达的动态改变。结果高氧后1 d,AEC-Ⅱ细胞器形态出现变化,随时间延长,细胞结构和形态发生严重改变,直至坏死;实验组肺损伤SP-C 3 d表达明显减少,7 d后开始增多,14 d、21 d明显高于对照组(P<0.05)。结论高氧肺损伤新生鼠肺泡AEC-Ⅱ形态结构明显改变,高氧严重影响了AEC-Ⅱ的部位,引起AEC-Ⅱ向AEC-Ⅰ转化障碍。Objective To explore the effect of hyperoixa on the morphological apperance,number and function of alveolar epithelial type Ⅱ cell(AEC-Ⅱ)in a neonatal rat model of chronic lung disease(CLD).Methods CLD was induced by hyperoxia.Eighty neonatal Wistar rats were divided to control group and model group randomly.Ultramicro-architecture of the lung tissues were observed by transmission electron microscope.Protein and mRNA expression of surfactant protein C(SP-C)were measured by immunohistochemisty and reverse transcription polymerase chain reaction (RT-PCR) respectively. Result Shapes of organelles in AEC-Ⅱ were changed from the 1st day in Model group. Cellular structures and morphological appearance were greatly altered into final necrosis with the exposure time prolonged. The expres- sion of SP-C was reduced on the 3rd in Model group and increased on the 7th day with significant differences observed on the 14th and 21st days between two groups (P 〈 0.05 ). Conclusion Structures and morphological appearance of AEC-Ⅱ were obviously altered on Model group. AEC-Ⅱ cells were seriously damaged during the hyperoxia exposure and finally result in their disturbed transformation into AEC-Ⅰ .
