摘 要:目的观察微囊化兔坐骨神经组织细胞移植对脊髓半横断后脊神经节P物质(SP)表达的变化。方法将实验动物分为对照组、损伤组、细胞悬液组和微囊组。免疫组织化学方法结合图像分析检测各组脊神经节SP的阳性神经元数和平均光密度值,并计算阳性细胞百分率。结果脊髓半横断损伤后第1、2周脊神经节内SP阳性神经元数、阳性细胞百分率、平均光密度值均降低,至第4、8周时逐渐回升。微囊组SP的表达明显高于同时间点的损伤组、细胞悬液组。细胞悬液组与损伤组比较,差别无统计学意义。结论微囊化兔坐骨神经组织细胞移植能使SP的表达上调。微囊化兔坐骨神经组织细胞移植可能通过使SP的表达上调在一定程度上促进其神经功能的恢复。Objective To observe the effects of microencapsulated rabbit sciatic nerve tissue transplantation on substance P(SP)expression in dorsal root ganglia(DRG)after spinal cord hemisection injury(hSCI).Methods The experimental rats were divided into 4 groups:the control group,the purely injured group,nervous tissue cells implanting group and microencapsalated nervous tissue cells implanting group after spinal cord right hemisection injury.Immunohistochemical staining and image analysis were used to detect the number of the positive neuruns and the mean optical density and the percentages of SP positive cells in the DRG. Results After hSCI, the number of the positive neurons and the mean optical density and the percentages of SP were reduced on the first and the second week. However, there was a gladU- al increase on the 4th and 8th week. Compared with nervous tissue cells implanting group or the purely injured group, all the parameters of SP were significandy higher in microeneapsalaled nervous tissue ceils implanting group at different time points. There was no significant change between nervous tissue cells implanting and the purely injured groups. Conclusion The xenotransplantation of microencapsulated rabbit sciatic nerve tissue cells can increase SP expression and consequently promote the recovery of nervous function.
