摘  要:目的了解单核细胞(Mo)组织因子(TF)表达在急性心肌梗死(AMI)和急性缺血性脑卒中(AIS)发作期间的变化,并观察它们是否存在差异。方法(1)采用半定量逆转录-聚合酶链反应(RT-PCR)法检测了20例AMI,20例AIS患者及61名正常对照者外周血Mo TF mRNA的表达;(2)采用发色底物法和ELISA法分别检测40例AMI和46例AIS患者血浆TF活性和TF抗原的水平,并与84名年龄匹配的健康对照比较。结果(1)与对照组相比,Mo TFmRNA表达在AMI及AIS组明显增高。(2)与对照组相比,血浆TF活性在AMI组增高,但无统计学差异,TF抗原显著增加;血浆TF活性及抗原在AIS组明显增加。(3)血浆TF抗原与Mo TF mRNA表达明显相关。结论(1)外周血MoTFmRNA在急性心脑血管血栓性疾病中呈高表达。(2)AMI和AIS均有组织因子途径的启动,其中TFAg显著增高是其共同特征。Objective To investigate the changes of tissue factor (TF) expression of monocytes (Mo) during the onset of acute myocardial in farction (AMI) and acute ischemic stroke(AIS). Methods ( 1 ) The expressions of Mo TF mRNA in 20 AMI patients, 20 AIS patients and 61 normal controls were tested, semiquantity reverse transcriptase- polymerase chain reaction ( RT- PCR) was used. ( 2 ) By means of chroMogenic substrate and ELISA, the levels of TF act and TFAg in plasma were detected in 40 AMI and 46 AIS patients at onset phase, 84 age-matched healthy volunteers respectively. Results (1)Compared with the control group ,the expressions of Mo TFmRNA were significantly higher in AMI and AIS groups. (2)Compared with the control group, plasma TF activity was higher in AMI, but not statistically difference, but TF antigens was significantly higher in AMI group; plasma TF activity and antigen in AIS group were significantly increased. (5) TF activity and TF antigen were significantly associated with the expression of Mo TF mRNA. Conclusions ( 1 ) The expression of TF mRNA in Mo was upregulation during the onset of AMI and AIS. (2) During the onset of AMI and AIS, the initiation of tissue factor pathway was associated with the cerebrocardiac thrombotic events and that the high level of TF antigens in plasma was the com Mo feaure.
