摘 要:目的:建立固体复合酶活力测定方法及采用固体复合酶测定果糖二磷酸钠(FDP)含量的酶反应方法。方法:在固体复合酶作用下,通过测量还原型辅酶Ⅰ(NADH)在340nm处吸收度的变化来测定果糖二磷酸的含量。结果:果糖二磷酸钠在0.1~0.6mg·mL-1范围内线性关系良好(r=0.9997),注射用果糖二磷酸钠平均回收率为99.3%(n=9);果糖二磷酸钠注射液平均回收率为99.0%(n=9)。结论:本法方法简便、准确,专属性强,可用于果糖二磷酸的含量测定。Objective:To establish a method for the determination of enzymatic activity of solid compound enzyme and established a method of enzymatic reaction for the determination of sodium fructose diphosphate(FDP) by solid compound enzyme. Method:The solid compound enzyme was used in the determination of sodium fructose diphosphate by absorption change of NADH at 340 nm. Result:There was a good linear relationship of sodium fructose diphosphate within the range of 0. 1 - 0.6 mg ·mL^-1 ( r = 0. 9997 ). The average recovery of sodium fructose diphosphate for injection was 99.3% ( n = 9). The average recovery of sodium fructose diphosphate injection was 99.0% ( n = 9). Conclusion : The method was simple, accurate, specific and suitable for the determination of sodium fructose diphosphate.
