摘  要:目的:建立HPLC-ELSD法测定人工蛹虫草子实体中甘露醇的含量。方法:采用氨基柱(4.6mm×250mm,5μm),柱温25℃,进样量20μL,以乙腈-水(82.5∶17.5)为流动相洗脱,流速0.5mL·min-1;ELSD漂移管温度90℃,载气(N2)流速2.0L·min-1,撞击器:关。结果:不同来源的人工蛹虫草子实体中甘露醇含量差别较大,其中以广东Ⅰ号厂家培育的含量最高(7.5%)。结论:本方法稳定、可靠,甘露醇可作为评价人工蛹虫草子实体质量的指标之一。Objective: To establish a reliable and accurate HPLC - ELSD method for the determination of mannitol in cultured Cordyceps militaris L. Link by optimizing the extraction, separation, and analytical conditions. Method: Separation was achieved on a NH2 column (4.6 mm×250mm,5μm) with acetonitrile - water ( 82.5: 17.5 ) as mobile phase at 25 ℃ and a flow rate at 0.5 mL ·min ^-1. The drift tube temperature of ELSD and nitrogen flow rate was set at 90 ℃, and 2.0 L· min ^-1, respectively, Impact : OFF. Results: Mannitol was found in 9 samples collected in different areas in China and the variety of rnannitol was significant. The highest content (7.5%) was found in Guangdong province. Conclusion: The current method demonstrated good linearity, selectivity, precision, recovery, short analysis time, and was successfully used to determine the content of mannitol in cultured Cordyceps militaris growing in different areas of China.
