花生去子叶胚离体培养时有关生理指标的变化

摘  要:将花生(Arachis hypogaea L.)粤优1号去子叶胚接种在以B5+0.20 mg/L 6-BA+0.20 mg/L NAA的培养基上,测定胚的萌发及幼苗各器官多胺氧化酶(PAO)、过氧化物酶(POD)的活性变化以及可溶性蛋白质含量和木质素含量的变化.结果表明,在萌发过程中,开始吸胀而未萌发的花生胚无PAO活性,直至2 d后才检测到活性,并且有一个峰值;而POD活性、蛋白质含量和木质素含量从胚吸胀第2 d就存在较高的活性,以后在第8 d^14 d降到峰底又开始回升,而这一低峰值被认为是植物从胚萌发到植株形态建成,从异养到自养的转折点.The decotyledoned peanut (Arachis hypogaea L. ) embryos of Yueyou No. 1 were in vitro cultured on Gmborg (B5) media supplemented with 6-BA 0. 20 mg· L^-1 and NAA 0. 20 mg· L^-1. Variation of the activities of polyamine oxidase (abbreviated as PAO), peroxidase (abbreviated as POD), and the contents of soluble protein and lignin in the decotyledoned embryos were determined during in vitro culture. The results indicated that there were no activities of PAO in the embryos at the first day of imbibed and ungerminated embryos. At the second day of imbibition, high activities of PAO and POD, and high contents of protein and lignin were detected, These physiological indexes decreased to the lowest level during the 8^th and 14^th day of culture, then began to increase again. The lowest level of these indexes may reveal the transition point from the heterophytic phase to the autophytic phase of the seedling growth and development.
