摘  要:目的探索构建经济实用的白色念珠菌性阴道炎动物模型的最佳成模条件。方法采用SPF级昆明雌性小鼠,经不同剂量的苯甲酸雌二醇(E2)处理后,小鼠阴道内注入含不同菌量白色念珠菌悬液造模。造模后动态观察小鼠的一般情况及外阴阴道症状,阴道灌洗液菌丝生成情况及菌落计数,并观察白色念珠菌性子宫积液情况及组织病理学情况。结果随着接种菌量及E2量的增加,阴道炎感染率随之增加,但白色念珠菌性子宫积液率亦增加。J3E2组(接种菌量为5×104个/只,皮下注射E20.025mg/只),第2天始即出现高水平的菌落数,CFU≥104/ml,第4天菌丝检出率可达80%,并可持续至实验结束,与接种菌量≤5×104个/只及皮下注射E2≤0.025mg/只的各组菌落数比较差异均有统计学意义(P<0.01或0.05),与接种菌量≥5×104个/只及皮下注射E2≥0.025mg/只的各组菌落数比较,差异均无统计学意义;且子宫积液率为33.33%,明显低于接种菌量≥5×104及皮下注射E2≥0.025mg/只的各组。感染成功小鼠阴道组织切片PAS染色阴道浅层组织及腔内见大量孢子及菌丝。结论J3E2组为昆明小鼠白色念珠菌性阴道炎模型最佳成模条件。Objective To search for optima construction conditions of economic model of Candida albieans vaginitis. Methods Four levels of the dose of estrogen benzoate(E2)was given to Kunming-mice from beginning 96 h to the end of the experiment. Vaginal inoculation was done with five levels of concentration of stationary-phase C. albicans blastoconidia. General state, genital pathology scores, the hyphae in vaginal lavage fluid, the vaginal fungal burden,the fluidify rate of uterus and vaginal tissues for pathological section were obtained after inoculation. Results The higher dose of estrogen benzoate injected hypodermically and more C. albicans blastoconidia inoculated intravaginally, the higher infection rate the mice would be, also the higher fluidify rate of uterus the mice would be. There was high load of C. albicans in the culture of lavage fluid(≥10^4/ml)from day 2 to day 21 after inoculation and high detection rate of hyphae(≥80%)from day 4 in group of J3E2(inoculated intravaginalty with 5 5〈 104 C. alhicans blastoconidia,0.025 mg E2 each mouse). There was significant difference when J3E2 compared with groups treated with less than 5× 10^4 C. albicans blastoconidia and 0. 025 mg E2 (P〈0.01 or 0.05) ,but there weren't significant difference when compared with the groups with more than 5 × 10^4 C. albicans blastoconidia and 0. 025 mg E2. The fluidify rate of uterus was about 33. 330/00 which was much lower than the groups with more than 5 × 10^4 C. albieans blastoconidia and more than 0. 025 mg E2. There were large amount of hyphae and blastoconidia by PAS stain in superficial layer tissue and canal of vaginal of the mice with vaginal candidiasis. Conclusion Group J3E2 was the optima construction condition in kunming-mice.
