摘  要:目的:研究神经再生素(NRF)对离体大鼠心肌缺血再灌注损伤的保护作用及机制。方法:建立Langendorff离体心脏体外灌注模型,观察心脏血流动力学变化:心脏左室发展压(LVDP)、左室舒张末压(LVDEP)、冠脉流量(CF)等心功能指标,测定灌流液中肌酸激酶(CK)含量,心肌组织中超氧化物歧化酶(SOD)和丙二醛(MDA)含量,光镜及电镜观察心肌结构及超微结构变化。结果:(1)NRF各用药组,LVDP较模型组明显升高(P<0.05,P<0.01),LVEDP则明显下降(P<0.01),CF值较模型组显著上升(P<0.01);(2)CK、MDA含量用药组较模型组明显下降(P<0.01),SOD活性用药组较模型组明显上升(P<0.01);(3)光镜结果显示用药组心肌结构较模型组明显好转,电镜显示模型组细胞超微结构损伤明显,用药组明显改善。结论:NRF具有明显的抗缺血再灌注损伤,保护心肌作用,其机制可能与其抑制心肌脂质过氧化损伤有关。OBJECTIVE To study the protective effect of NRF on ischemia/reperfusion injury of isolated rats hearts. METHODS Langendorff retrograde perfusion was performed as described by feinberg. The following indexes of rats in each group were measured respectively: left ventricular developed pressure(LVDP), left ventricular end diastolic pressure (LV EDP), Creatine kinase (CK) leakage, the contents of superoxide dismutase(SOD), malondialdehyde (MDA). Ultra structure of the cardiac muscle in the left ventricle was examined using a transmission eletron microscope. RESULTS (1) Compared with 1/R group, I.VDP was increased significantly in I/R + NRFm group and I/R + NRFI group while the LVEDP was lowered significantly (P〈0. 01 ,P〈0. 05). (2) NRF relieved the injury: the activities of SOD was significantly higher, CK and MDA con tents were significantly lower than those in 1/R group. (3) Most cells of I/R group by electron microscopy appeared as the necrosis, When the hearts treated with NRF the condition were ameliorated.
