摘 要:目的:应用细胞生物学技术探讨红景天多糖(RSP)体外抗柯萨奇B组3型病毒(CVB3)活性和对心肌细胞保护作用机制,探究其作用环节(药物靶点),为RSP的开发利用奠定基础。方法:VERO细胞感染CVB3病毒观察细胞病变;CVB3病毒感染乳鼠心肌细胞建立病毒性心肌炎细胞模型,分析RSP对VERO和心肌细胞的保护作用;病毒繁殖抑制实验验证RSP在VERO细胞及心肌细胞上的抗病毒活性;紫外分光光度法测定乳鼠心肌细胞培养上清液中乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK-MB)的活性;流式细胞术和RT-PCR检测比较各组间细胞凋亡的差异。结果:RSP在VERO细胞和心肌细胞试验中可有效抑制CVB3病毒引起的细胞病变(P<0.05,P<0.01);且对病毒的繁殖有抑制作用(对数值升高一个以上数量级),降低心肌酶(LDH、CK-MB)的释放,对细胞凋亡有抑制作用(P<0.01)。结论:RSP对CVB3病毒感染的VERO细胞和原代乳鼠心肌细胞具有一定的保护作用。对CVB3病毒的繁殖有抑制作用,在一定浓度下可降低心肌酶的释放并抑制细胞凋亡,其作用机制尚需做进一步试验来探讨。OBJECTIVE To study the effect of the RSP(Rhodiola sachalinensis polysaccaride A)and explore the antiviral mechanism of the RSP on the model of viral myocarditis in vitro. METHODS Cytopathie effect(CPE) was observed with inverted phase contrast microscope and cell viability was calculated. Cardiac myocytes derived from newborn Sprague-Dawlay rats was identified. To investigate RSP'inhibition of CPE and infectious virus propagation about VERO and cardiac myocytes infec ted by Coxsackievirus B3 (CVB3). Activity of 50% tissue culture infective dose(TCID50 ) was used to evaluated the infectious effect of CVB3. Derived from the supernatants cultured VERO cell infected by CVB3 preciously. At the same time, supernatants of cultured cardiac myocytes were collected to test the activity of LDH,CK-MB in different groups. RESULTS The CPE of VERO and cardiac myocytes were alleviated by RSP in vitro (P〈0. 05, P〈0. 01 ). RSP inhibited the propagation of CVB3 and decreased the activity of LDH, CK-MB when it was compared with that of model Group(P〈0.05 ,P〈0. 01 ). CONCLUSION The VERO and cardiac myocytes infected by CVB3 were protected by RSP. RSP inhibited the propagation of and decreased the activity of LDH, CK-MB released from cardias mycytes. The mechanism of the RSP will be studied in the future.
