摘 要:系统地研究了瓠瓜子叶外植体直接发生不定芽,以及不定芽生根壮苗培养的各种影响因子.实验结果表明:不同部位的子叶外植体其不定芽发生的频率高低依次为子叶基段>子叶中段>子叶顶段;具有较多创口的外植体(打孔或切开伤口)其不定芽发生频率较高;6种供试基本培养基用于诱导瓠瓜子叶直接发生不定芽的效果依次为MS>1/2 MS>1/3 MS>B5>N6;在KT、6-BA和ZET等3种外源细胞分裂素的不同质量浓度处理中,以1.0 mg.L-1 6-BA的处理对于诱导瓠瓜子叶外植体直接分化出不定芽有比较理想的效果;VB1的效果不明显;VB6有较好的促进作用,其中以附加0.5 mg.L-1 VB6的处理效果最好(不定芽发生率53.3%).附加1.0 mg.L-1 NAA的MS培养基用于不定芽的生根壮苗培养可获得健壮的完整植株,其根系发育良好,平均单株根数达5.3条.The various effective factors, which effected the cotyledon explants directly from adventitious buds, and that the promoting-rooting-strengthening-plantlet culture of the shoots, were systematically studied. The experimental results showed that the frequency sequence of the adventitious buds of the explants which were located at the different parts of the cotyledon was as the follows: The basic segment of cotyledon 〉 the middle segment of cotyledon 〉 the top segment of cotyledon. These explants with more wounds (by punching or by cutting) had a higher frequency of the ad- ventitious bud direct formation. The effect order of the 6 test basic media, used for the inducement of the direct formation of adventitious buds was as follows: MS〉3/2 MS〉 1/3MS 〉 B5 〉 N6. Of all 3 kinds of test exogenous cytokinin (KT, 6-BA and ZET) with different concentrations, the treatment complemented with 1.0 mg· L^-1 6-BA showed a fairly good effect for inducement of adventitious buds directly from the cotyledon explants. VB, showed a unclear effect on the inducement of direct formation of the adventitious buds from the cotyledon explants, and VB6 showed a fairly good effect on the direct formation of the adventitious buds. Of all VB6 concentration treatments, the treatment with 0.5 mg·L^-1 VB6 was the best (The formation frequency of the adventitious buds was 53.3 %). Medium MS complemented with 1.0 mg· L^-1 NAA showed ideal effect for the promoting-rooting-strengthening-plantlets culture. The robust complement plants, with well developed footage, could be obtained on the MS complemented with 1.0 mg ·L^-1 NAA. The single plantlets had 6.3 roots, meanly.
