摘 要:以花楸(Sorbus pohuashanesis(Hance) Hedl)幼胚为外植体,对不定芽诱导、增殖和腋芽增殖过程中玻璃化现象的发生和防治措施进行了研究。结果表明:培养基种类和激素质量浓度是导致玻璃化现象的2个主要原因。在花楸组织培养中MS和WPM是理想的培养基;在不添加任何激素的MS和WPM培养基上,添加琼脂7g/L,并用带透气孔的封口膜封口,绝大部分玻璃化苗可逆转为正常的健壮苗,而且在WPM培养基上,玻璃化苗在逆转为正常苗的同时有60%~80%的茎芽基部长出辐射状根。An experiment was conducted to study the vitrification and its prevention during the induction of indefinite buds, mul- tiplication of adventitious shoots from young embryos, and the proliferation of auxiliary buds on the in vitro cultured shoots of Sorbus pohuashanensis (Hance) Hedl. Results indicated that medium type and hormone concentration were two important factors to cause vitrification. MS and WPM were the optimal media for tissue culture of S. pohuashanensis. Almost all of the vitrified shoots could become vigorous with usual appearance on hormone-free MS or WPM medium supplemented with 7 g/L agar in culture containers with the ventilation hole sealed with sealing film. Moreover, 60 percent to 80 percent of shoots grew radial roots at their bases on WPM medium during the reversion period from vitrification to normal.
