摘 要:用结晶紫染色法^3H-TdR掺入,增殖细胞核抗原免疫化学法检测3,4-二羟基苯乙酮对缺氧所致独肺动脉平滑肌增殖的影响。结果发现缺氧条件下培养的PASMC用以上3种方法测得结果分别为常氧组的。The effect of 3, 4-dihydroxyacetophcnone (DHAP) on the proliferation of the cultured pulmonary arterial smooth muscle cells (PASMC) induced by hypoxia was studied with methods of crystal violet staining. 3H-TdR incorporation and proliferating cell nuclear antigen (PCNA) immunocytochemistry. The results showed that the data detected by these three methods in the hypoxic PASMC group were (117. 1±4. 7) %, (131.2±17.0) %, (112.8±6. 2) % respectively, as much as those in the normoxic group (P<0.01 for all), which implied that hypoxia 3,4-PHAP could inhibit the proliferation of PASMC. induced by hypoxia in a dose-dependent manner. Since the hyperplasia of SMC contributes to the remodeling of pulmonary vessels in chronic hypoxic pulmonary hypertension, 3,4-DHAP might be of benefit in its prevention and treatment.
