摘 要:目的观察IL-17在日本血吸虫感染小鼠中对抗体及Th细胞免疫应答的影响。方法在小鼠日本血吸虫感染前后,连续多次腹腔注射rm-IL-17细胞因子(实验组1)或PBS(对照组1),连续多次腹腔注射anti-IL-17中和抗体(实验组2)或同型抗体(对照组2)。运用流式细胞术,采用表面分子、胞内因子同时染色的三色标记法,对日本血吸虫感染小鼠脾脏、淋巴结中的Th17、Th1和Th2细胞亚群占CD4+T细胞的比例进行观察;同时采用ELISA法检测各组小鼠血清中血吸虫抗原特异性总IgG及亚类IgG1和IgG2a的水平。结果实验组1 CD4+T细胞中的Th17、Th1和Th2细胞亚群的比例分别是对照组1的1.09倍(P(0.05)、1.17倍(P(0.05)和1.15倍(P(0.05);实验组2 CD4+T细胞中的Th17、Th1和Th2细胞亚群的比例分别是对照组2的1.01倍(P(0.05)、1.11倍(P(0.05)和0.98倍(P(0.05)。rm-IL-17注射后仅可溶性虫卵抗原(SEA)特异性IgG1水平显著下降(P(0.05)、可溶性成虫抗原(SWA)特异性IgG2a的水平升高(P(0.05),其他抗体水平均无显著变化;anti-IL-17中和抗体注射后,SEA特异性总IgG(P(0.05)、IgG2a(P(0.01)和SWA特异性IgG2a(P(0.01)水平显著升高,其他抗体水平均无显著变化。结论IL-17可能不会显著增强或降低宿主的血吸虫特异性Th细胞应答,而可能对IgG类抗体的产生具有某种程度的抑制作用。Objective To observe the antibody and Th responses affected by IL-17 in mice infected with Schistosomejaponicum. Methods By repeated administration of rmIL-17 ( test group 1 ) or PBS ( control group 1 ), or anti-IL-17 neutralizing mAb ( test group 2 ) or its IgG2a isotype control antibody ( control group 2 ) before and after Schistosoma japonicum infection, the proportions of Thl7, Thl and Th2 populations in C57BL/6 mouse spleen and lymph nodes were stained with surface molecule and intracellular cytokines and examined by three-color flow cytometery. Meanwhile, the levels of schistosome antigen specific IgG, IgG1 and IgG2a antibodies in mouse sera were detected by ELISA. Results In the rmIL-17 injected group, the proportions of Th17,Thl and Th2 populations in CD4^+T cells were 1.09 folds (P〉0.05) , 1.17 folds (P 〉 0.05) and 1.15 folds (P 〉 0.05 ) of those in PBS injected control group, respectively. In the anti-IL-17 neutralizing mAb injected group, the proportions of Thl7,Thl and Th2 populations in CD4*T cells were 1.01 folds (P 〉 0.05), 1.11 folds (P 〉 0.05) and 0.98 folds (P 〉 0.05 ) of those in the IgG2a isotype antibody injected control group, respectively. In the rmIL-17 injected group, SEA spcific IgGl significantly decreased ( P 〈 0.05 ) and SWA specific IgG2a slightly increased ( P 〈 0.05 ), while the other antibodies did not change significantly. In the anti-IL-17 neutralizing mAb injected group, SEA specific IgG (P 〈 0.05 ), IgG2a (P 〈 0.01 ) and SWA specific IgG2a (P 〈 0.01 ) significantly increased,but the other antibodies did not change significantly. Conclusion IL-17 might not "affect the Th response but down-regulate the production of antibodies during Schistosoma japonicum infection, and the related mechanism needs further illustration.
