摘  要:目的观察日本血吸虫钙离子通道变异的β亚基(SjCavβvar)在成虫期和童虫期转录本的表达情况,为研究吡喹酮的作用机制提供依据。方法根据日本血吸虫特异性基因序列,设计实时荧光PCR特异性引物,采用SYBRGreen染料法检测SjCavβvar2个虫期的转录本水平。结果雄虫、雌虫与童虫SjCavβvar的mRNA水平差异无统计学意义(P>0.05)。结论SjCavβvar参与吡喹酮作用的机制可能与其他亚基的共表达有关。The transcript levels of schistosome voltage-gated calcium channel variant 13 subunit (SjCavβvar) in the adult worm and schistosomulum stages were observed, so as to provide the evidence for the study of praziquantel (PZQ) action mechanism. According to the specific sequence of schistosome, the specific primers of real time PCR were designed and SYBR Green I dye was used for detecting the transcript levels of SjCavβvar in the two stages. The results revealed that there were no significant differences of the expression levels of mRNA between the two stages. It is suggested that the involvement of SjCavβvar in the mechanism of action of PZQ may be related to the co-exoression with other subunits.
