摘　　要：为了探索鸡分泌型免疫球蛋白A（SIgA）分离、纯化及鉴定方法,以新鲜的鸡胆汁为材料,采用硫酸铵盐析法从鸡胆汁中粗提纯鸡SIgA,应用SDS-聚丙烯酰胺凝胶电泳（SDS-PAGE）鉴定和纯化SIgA重链,用纯化的鸡SIgA重链蛋白免疫Balb/C小鼠。结果表明,SIgA重链分子量为67-70kD,轻链为26-28kD。免疫Balb/C小鼠抗血清针对SIgA重链的间接ELISA效价可达1∶16000。为进一步研究SIgA生物学特性、单克隆抗体制备和黏膜免疫状况检测提供了技术支持。The separation, purification and identification of the chicken secretary immunoglobulin A（SIgA） were carried out. Using improved method of ammonium sulfate, the chicken SIgA was extracted from the fresh galls. The heavy chain of SIgA was purified by sodium sulphate polyacrylamide gel electrophoresis （SDS- PAGE） and was used for the immunization of Balb/C mice. The results indicate that the molecular weight of the heavy chain and light chain were about 67-70kD and 26-28kD. The titers of antisera prepared from the mice reached to 1 ： 16000. This provides technical supports for deeply researching on the biological characteristics of SIgA, preparation of the monoclonal antibody and detection of the situation of mucosal immune.
