摘  要:目的:探讨三七总苷(TNG)对5/6肾切除大鼠慢性肾衰竭(CRF)模型的肾脏保护作用及其可能机制。方法:SD大鼠60只,随机分为假手术组和手术组,假手术组再随机分成假手术未治疗组和假手术TNG治疗组;手术组行5/6肾切除术后3周确定造模成功后再随机分为模型未治疗组、模型TNG低、中、高剂量组。术后第5周开始灌胃,治疗4周后留取24 h尿液检测24 h尿蛋白排泄量,后处死大鼠。全自动生化分析仪检测大鼠血清肌酐、血尿素氮、血红蛋白和电解质;免疫组织化学法和RT-PCR法检测各组大鼠肾组织纤维连结蛋白(FN)及mRNA的表达。结果:TNG能显著改善CRF大鼠一般情况,降低24 h尿蛋白排泄量,降低血清肌酐和尿素氮水平,纠正贫血及电解质紊乱,减少FN在肾组织中的表达,且呈剂量依赖趋势。结论:TNG可能通过下调肾组织细胞外基质的合成而对5/6肾切除大鼠残肾功能起保护作用。AIM: To explore the protective effect of total notoginsenosides (TNG) on chronic renal failure (CRF) rat model and its possible mechanism. METHODS : Sixty SD rats were randomly divided into sham operation group( including untreated group and TNG therapy group) and operation group which was further divided into operation untreated group, TNG low, medium and high dose therapy groups. Then the rats in TNG therapy groups received TNG therapy delivered by gavage for 4 weeks, while the rats in untreated groups only received distilled water since 5 th week of operation 24 h urinary protein excretion (24 hUPE) as well as levels of Scr, BUN, Hb and electrolytes were detected after 4 weeks of treatment. Renal expression of fibronectin(FN) was detected by immunohistochemical staining and RT-PCR. RESULTS: TNG can markedly improve rat's general state, lower 24 h UPE and serum creatinine and urea level, correct anemia and electrolyte disturbance, decrease FN expression in kidney, and has a great liability to the dose-effect relationship. CONCLUSION: TNG can protect residual renal function in CRF rats. And the possible mechanism is to down-regulate renal expression of FN.
