摘  要:目的探讨血脂康对氧化型胆固醇(Ch-Ox)中25-羟胆固醇(25-OH)、3β,5α,6β-三羟胆固醇(3-Triol)导致体外培养胰岛素细胞活性损害的保护及抗凋亡作用,并观察其量效关系。方法体外培养小鼠胰岛素细胞株MIN6细胞,待细胞生长融合后,加入50μmol/L浓度的Ch-Ox(3-Triol与25-OH)和不同浓度的血脂康,共同孵育24h。采用MTT法检测细胞的存活情况,并用TUNEL法检测胰岛细胞凋亡率。结果Ch-Ox可降低MIN6胰岛细胞活性,增加MIN6胰岛细胞凋亡率;加入血脂康液后,对Ch-Ox导致的MIN6胰岛细胞活性降低起保护作用,且呈剂量依赖性,有明显统计学差异;另外,血脂康液剂量依赖性减少MIN6胰岛细胞凋亡率。结论血脂康对Ch-Ox导致体外培养胰岛素细胞活性的损害具有保护作用及抗凋亡作用,并存在剂量依赖效应。Objective To investigate the protective and antiapoptosis effect of Xuezhikang in islet cells damged by Oxysterols (25-OH or 3-Triol) ,and observe the dose-effect relationship. Methods MIN6 mouse islet cells were cultured in vitro. After confluence of cells,50 μmmol/L Oxysterols (3-Triol or 25-OH ) was added to culture medium to act for 24 hours, on the other hand, different dose Xuezhikang was added to culture medium to act for the same time. MTT test was used to evaluate the viability and TUNEL test was used to detect the apoptosis of MIN6 cells. Results Oxysterols (25-OH or 3-Triol) decreased the viability of MIN6 insulin-secreting cells, and induced the apoptosis. There was significant difference between viability of islet cells with Xuezhikang as a protective effort. While a decrease in apoptosis cells was observed in a dose-dependent and time-dependent manner treated with Xuezhikang. Conclusion Both 3-Triol and 25-OH can induce apoptosis and decrease the viability of MIN6 insulinsecreting cells, while the Xuezhikang has a protective effect on viability of islet cells and anti-apoptotic function in a dose dependent manner.
