摘 要:目的探讨别嘌呤醇对心肌缺血再灌注大鼠左室重构的影响。方法Wistar大鼠24只,雌雄各半,随机分为假手术组、缺血再灌注组(MIR组)及别嘌呤醇组(别嘌呤组),每组8只。制成大鼠心肌缺血再灌注模型。每组连续灌胃4周,将别嘌呤醇稀释后按10 ml/kg,30 mg/kg,灌胃,假手术组、缺血再灌注组均以等量生理盐水灌胃。测定以下指标:(1)心室大体、光镜、电镜变化:左室相对重量、室间隔厚度。(2)生化指标:测定血清中丙二醛(MDA)、超氧化物歧化酶(SOD)含量。结果(1)大体标本:MIR组与假手术组比较,左室相对重量、室间隔厚度显著增加;别嘌呤组与MIR组比较,左室相对重量显著减轻,室间隔厚度显著减少。(2)光镜结果:假手术组心肌纤维排列规整,心肌细胞未见坏死,心肌间质无水肿;MIR组心肌纤维排列紊乱,心肌细胞可见坏死,心肌间质明显水肿。别嘌呤组:心肌纤维略增粗,排列较规整,心肌细胞可见少量坏死,心肌间质无明显水肿。(3)电镜结果:假手术组肌丝束排列整齐,肌节明暗带清晰可见,线粒体未见异常;MRI组:肌丝束局部有断裂和溶解,线粒体局部有空化改变;别嘌呤组:肌丝束排列尚整齐,肌节明暗带尚清晰,局部线粒体空化改变减少。(4)生化指标:MIR组与假手术组比较,大鼠MDA含量明显升高,SOD含量明显降低;别嘌呤组与MIR相比,MDA含量显著降低,SOD含量升高。结论别嘌呤醇能有效改善MIR大鼠的左室重构,其机制可能与别嘌呤醇清除氧自由基有关。Objective To observe the treatmentation of Allo on myocardial ischemia reperfusion models, the aim is to explore the effection of Allo on left ventricular remodeling in myocardial ischemia reperfusion (M1R) rats. Methods Rats were randomly divided into sham group; MIR group and Allo treatment.group. While models are successfully producted, then medicines are administrated through garage for at least four weeks. We observe the left ventricular remoding from 3 directions specimen; light microscopy; electron microscopy, and then by testing serum superoxide dismutase (SOD), malondialdehyde (MDA), and other indicators. Results First, ventricular remodeling indicators contrast results:Allo group compared with the Mill group, left ventricular relative weight and septal thickness are significantly reduced. Seeond,light microscopy results:sham group myocardial fiber,myocardial ceils and myocardial interstitial are no obvious abnormalities. Myocardial fiber of Mill group arrange disorder. Myocardial cells shows necrosis, myocardial interstitial are obviously edema, Ailo group than MIR group is to reduce ventricular remodeling. Third, electron microscopy results: myofilament, sarcomere and mitochondria of sham group are no obvious abnormalities. Muscle tow of MIR group have fracture and dissolution partly. The mitochondria of MIR group become smaller, and have cavitation. Myofilament of 331o group arrange in order, sarcomere shad with clear, and have cavitation. Forth, biochemical indicators comparative results: There are less MDA in Allo group than MIR group,but more SOD in Allo group than MIR group. Conclusion Allo can be effective in improving left ventricular remodeling, which can remove free oxegen radicals.
