摘  要:[目的]研究鱼藤酮引起的星形胶质细胞内游离钙浓度变化及其离子流动机制。[方法]以体外培养的大鼠中脑星形胶质细胞为研究对象,激光共聚焦结合Fluo-4荧光法动态测定细胞内游离钙浓度。[结果]鱼藤酮可浓度依赖性地引起星形胶质细胞内游离钙浓度升高。细胞内钙库释放抑制剂TMB-8、无钙缓冲液和钙通道阻滞剂MK-801、尼莫地平均可抑制鱼藤酮引起的细胞内钙浓度变化,但其抑制作用发挥的时间和程度均有显著不同。[结论]细胞内钙释放与细胞外钙内流共同参与了鱼藤酮引起的星形胶质细胞内游离钙浓度的升高,但细胞内钙释放发挥作用的时间稍迟,而鱼藤酮引起的细胞外钙内流主要由电压依赖式钙通道内流引起。[ Objective] To investigate the intraeellular calcium concentration alteration induced by rotenone and the mechanism of ion flux in astroeytes. [ Methods] Rat meseneephal astmeytes were cultured in vitro. Intraeellular calcium concentrations were measured by laser scanning eonfocal microscope and Fluo-4 fluorescence dye technique. [ Results] Rotenone significantly increased intraeellular calcium concentration in a dose-dependent manner. Intraeellular calcium store released blocker (TMB-8), Ca^2+-free bath, and calcium channel blocker (MK-801 and nimodipine), which inhibited the elevation of intraeellular calcium concentration induced by rotenone, but the inhibiting time and level were also significantly different. [Conclusion~ Intraeellular Ca^2+ elevation induced by rotenone is due to both the release of intraeeUular calcium store and extracellular calcium entry, but the release of intraeellular calcium stores occurs in the latter phase. Rotenone-indueed extraeellular calcium entry is mainly through voltage-operated Ca^2+ channel.
