大鼠骨骼肌缺血再灌注损伤后促凋亡基因Bax蛋白的表达

摘  要:[目的]探讨大鼠骨骼肌缺血再灌注后不同时相肌细胞Bax基因蛋白表达的变化。[方法]用免疫组织化学方法检测并观察大鼠右后肢止血带缺血再灌注后4h、24h、48hBax基因蛋白在肌细胞中的表达情况;均以同组大鼠左后肢腓肠肌作自身对照。[结果]缺血再灌注后4h肌细胞内出现Bax蛋白表达,其阳性表达率为(5.97±1.44)%;缺血再灌注后24h,肌细胞Bax蛋白表达的阳性率为(18.45±1.81)%;缺血再灌注后48的肌细胞则未见Bax蛋白的表达;所有各组左后肢肌细胞内亦无Bax蛋白的表达。统计分析表明,缺血再灌注后24h组较之4h组、48h组以及实验自身对照组差异有统计学意义(P﹤0.01)。[结论]本实验研究表明,缺血再灌注损伤的肌细胞中有促进细胞凋亡基因Bax蛋白的阳性表达,其阳性表达率与缺血再灌注不同时相密切相关,以24h组为最显著,此时病理损害亦最为显著,提示Bax基因促发的细胞凋亡可能是骨骼肌缺血再灌注损伤病理机制中的一个组成部分。[ Objective] To examine the expression of a proapoptotic protein, Bax in the skeletal muscle at different phases in a rat ischemic/reperfusion model. [Methods] Tourniquet was used to produce the rat ischemic/reperfusion model in the right hindlimb and the left side served as control. The expression of Bax was examined in the gastrocnemius 4, 24 and 48 hours after the ischemic procedure using lmmunohistochemical method. [ Results] The positive expression rate of Bax was 5.97±1.44% 4 hours after the ischemic injury, this rate tripled at 24 hours (18.45%±1.81%), and had no expression at 48 hours. The expression of Bax at 24 hours was significantly higher than the other two time points, which was coincident with the greatest pathological change of the skeletal muscle. [ Conclusion] We conclude that there is expression of proapoptotic protein Bax after ischemic/reperfusion injury m the skeletal muscle, and its expression peaks at the time coinciding with the greatest pathological feature in the skeletal muscle, indicating that apoptosis contributes to the post-ischemic/reperfusion muscle injury.
