摘  要:研究小麦叶绿体DNA,需要快速,简便地获得质纯、量大的ctDNA.将高离子强度和抗坏血酸同时引入缓冲系统用于提取小麦ctDNA,利用高离子强度对DNA与蛋白质之间的静电吸引造成裹胁损失的阻碍作用,及抗坏血酸提供低pH环境抑制酶活性等作用,得到较好的产率和纯度,并可得到清晰的限制性内切酶图谱.The more effectively and more rapidly method was used to purify ctDNA of wheat in this paper.The high ionic strength and ascorbic acid were used to get higher yield and better purity.A clearly restriction enzyme mapping would be obtained.High ionic strenth would decrease the electrostatic interaction,and decrease sticking loss that was led by electrostatic interaction between the DNA and protein.Ascorbic acid would provide a low pH condition to restrain the activity of enzyme.
