摘  要:目的探讨大鼠液压冲击伤后水肿脑组织AQP4表达变化与脑水肿之间关系。方法成年雄性SD大鼠48只制作颅脑损伤模型,术后6、12、24h、3、7d用干湿重法、免疫组织化学法分别测定水肿脑组织含水量及AQP4表达。结果自伤后6h,脑组织含水量开始增加(78.48±0.50)%,24h达高峰(82.54±1.89)%,持续至3d(80.61±0.49)%,然后开始下降,至7d基本恢复到正常。组织学证实了脑水肿趋势。免疫组织化学显示水肿脑组织AQP4表达于伤后6h开始增加(0.178±0.016),24h达高峰(0.331±0.024),持续至3d后下降(0.307±0.019),7d仍高于假手术组;线性相关分析发现AQP4表达与脑含水量呈正相关(r=0.940,P〈0.05)。结论AQP4表达上调可能在鼠液压冲击伤脑水肿的消散中起重要作用。Objective To investigate the relationship between AQP4 expression and cerebral ede- ma following fluid percussion injury in rats. Methods The experimental models were established in 48 rats. The water content and AQP4 expression were determined with dry-wet measure, and immunohistochemistry at the 6th h, 12th h,24th h,3rd day, and 7th day after operation respectively. Results As compared with sham operated group, the water content was increased in model group from 6 h [ (78.48 ± 0. 50) % ], reached its peak at 24th h [ (82.54 ± 1.89) % ], lasted to 3 rd day [ ( 80.61 ± 0.49) % ], and then began to reduce, nearly to normal level at 7th day, while AQP4 immunoreactive expression increased from 6 h (0. 178 ±0.016) ,reached its maximum at 24th h (0.331 ±0. 024) ,lasted to 3rd day (0.307 ± 0.019) ,and began to reduce. Until the 7th day,AQP4 immunoreactive expression was still higher than that in sham operated animals. The linear regression analysis indicated that the expression of AQP4 in edematous brain had a positive correlation with change of brain water content ( r = 0.940, P 〈 0.05). Conclusion The up-regulation of the AQP4 expression may be involved in water clearance of cerebral edema following fluid percussion injury in rats.
